4E-BP2–dependent translation in parvalbumin neurons controls epileptic seizure threshold

چکیده
Significance In numerous models of neurodevelopmental disorders and epilepsy, genetic mutations associated with enhanced mTORC1 activity engender an increase in the susceptibility to epileptic seizures. This study shows that deletion mTORC1-downstream translational repressor, 4E-BP2, but not 4E-BP1, promotes epileptogenesis. By deleting 4E-BP2 different cell types, we reveal up-regulation mTORC1/eIF4E signaling parvalbumin inhibitory neurons is sufficient reduce threshold for induction We conclude downstream mTORC1, main 4E-BP paralog controlling epileptogenesis propose are a vulnerable type which aberrant activation mTORC1-dependent translation contributes pathology.

Seizure threshold in electroconvulsive therapy: I. Initial seizure threshold.
We measured initial seizure threshold by means of a structured stimulus dosage titration procedure in a clinical sample of 111 depressed patients undergoing brief-pulse, constant-current electroconvulsive therapy (ECT). Initial seizure threshold was approximately 60 millicoumbs (mc) (10 Joules) on average, but varied widely (6-fold) across patients. Initial seizure threshold was predicted by fo...
متن کاملAkinetopsia as epileptic seizure☆
Akinetopsia is a rare syndrome in which a patient specifically loses the ability to perceive visual motion following bilateral cortical lesions outside the striate cortex. We describe a patient who showed akinetopsia recurrently as epileptic seizures. The patient was a 61-year-old man. At age 46, a cerebral arteriovenous malformation in the right parietal lobe was discovered. At age 58, he bega...
متن کاملAlgesthesia after epileptic seizure.
The consequences of epileptic seizures related to postictal inhibition in early postictal period include postictal analgesia. We studied this phenomenon over 96 h following flurothyl-induced seizures in adult male Wistar rats. Nociception of control (no seizure) and seizured groups were tested using the plantar and von Frey hair tests. We determined latency of forepaw and hind paw reactions usi...
متن کاملEpileptic Seizure Prediction: An overview
Epilepsy is a neurological disorder marked by sudden recurrent episodes of sensory disturbance, loss of consciousness, or convulsions, associated with abnormal electrical activity in the brain. The sudden and seemingly unpredictable nature of seizures is one of the most compromising aspects of the disease epilepsy. Most epilepsy patients only spend a marginal part of their time actually having ...
متن کاملThe Translational Repressor Pumilio Regulates Presynaptic Morphology and Controls Postsynaptic Accumulation of Translation Factor eIF-4E
Translational repression by Drosophila Pumilio (Pum) protein controls posterior patterning during embryonic development. Here, we show that Pum is an important mediator of synaptic growth and plasticity at the neuromuscular junction (NMJ). Pum is localized to the postsynaptic side of the NMJ in third instar larvae and is also expressed in larval neurons. Neuronal Pum regulates synaptic growth. ...
